taking a sample from the male, dividing the sample and counting the sperm in a smaller portion, then multiplying to extrapolate the count to the full sample and know how many sperm are working on the job of fertilizing a female egg.
The second factor is the sperm quality. The fertility lab will judge the average activity and virility of the individual sperm, thus noting the energetic factor a man's sperm has of completing the fertilization. Numbers times energy equals the stimulation an egg receives to admit one simgle sperm inside for fertilization. Without an adequate amount of stimulation by the mass of sperm working on the egg, this will not happen and fertilization will have failed.
